40 weeks does equal 280 days, but fertilization occurs 14 days after the last period (in a 28 day menstrual cycle). Thus, the typical duration of a human pregnancy is 280-14 = 266 days. We rounded up to 270, which is the number often used in pregnancy calculations. Babies born before Day 270 are termed premature.
